P-3073 is an investigational topical nail solution containing 0.005% calcipotriene (also known as calcipotriol), developed by Polichem S.A. (a subsidiary of Almirall) for the treatment of nail psoriasis. Calcipotriene is a synthetic vitamin D3 analog that acts as an agonist of the vitamin D receptor (VDR). Upon binding to the VDR, it modulates the expression of genes involved in keratinocyte proliferation and differentiation, effectively inhibiting hyperproliferation and inducing normal maturation of keratinocytes within the nail unit. It also possesses anti-inflammatory properties by modulating T-cell activity and cytokine production. P-3073 is specifically formulated to enhance penetration through the nail plate to reach the underlying nail bed and matrix. It has been evaluated in Phase 3 clinical trials for patients with mild to moderate fingernail psoriasis, including those with isolated nail involvement or concomitant plaque psoriasis.
