Overview

P-BCMA-101 is an autologous chimeric antigen receptor (CAR)-T cell therapy developed by Poseida Therapeutics using the piggyBac transposon-based gene modification system. It targets B-cell maturation antigen (BCMA), a protein highly expressed on multiple myeloma cells. Unlike traditional CAR-T therapies that use viral vectors, P-BCMA-101 employs a nonviral approach, utilizing only plasmid DNA and mRNA for gene transfer, which reduces manufacturing costs and increases the proportion of desirable stem cell memory T cells (Tscm). The therapy uses a fully human Centyrin binding domain fused to a second-generation CAR scaffold, designed to minimize immunogenicity and tonic signaling while enhancing stability and efficacy. Clinical studies have shown promising safety and efficacy in relapsed/refractory multiple myeloma, leading to FDA orphan drug designation for this indication[1][3][4][6][8].
