P-BCMA-ALLO1 is an off-the-shelf, allogeneic chimeric antigen receptor T-cell (CAR-T) therapy designed to target B-cell maturation antigen (BCMA). This therapy utilizes allogeneic T cells, rich in stem cell memory T cells (TSCM), which are genetically edited using non-viral methods to express a CAR that targets BCMA, a protein highly expressed on malignant plasma cells. The product is intended to replace a patient's dysfunctional T-cell population with functional cells capable of recognizing and destroying myeloma cells through immunologic cytotoxicity. It is developed for the treatment of relapsed/refractory multiple myeloma in patients who have received at least three prior lines of therapy. P-BCMA-ALLO1 is under development by Poseida Therapeutics in collaboration with Roche, and has received both orphan drug designation and Regenerative Medicine Advanced Therapy (RMAT) designation from the US FDA. The product is being evaluated in Phase 1/1b clinical trials, showing high response rates and a favorable safety profile without inducing dose-limiting toxicities, graft-versus-host disease, or severe neurotoxicity[1][2][3][4][5][6][7][8].
