Overview

**P-BCMACD19-ALLO1** is a preclinical, donor-derived, fully allogeneic dual-targeting CAR-T-cell therapy developed by Poseida Therapeutics and subsequently acquired by Roche. It is designed for relapsed or refractory multiple myeloma and targets both B-cell maturation antigen and CD19, including a CD19-positive multiple-myeloma progenitor population implicated in resistance to BCMA-directed therapy. The product is enriched for T stem cell memory cells and uses nonviral transposon-mediated gene delivery; TRBC1/TRBC2 disruption is intended to reduce graft-versus-host disease, while beta-2-microglobulin disruption is intended to reduce host T-cell-mediated rejection. Its multicistronic transgene encodes separate BCMA- and CD19-directed CARs, an inducible caspase-9 safety switch, and a dihydrofolate reductase mutein. Preclinical studies showed cytotoxicity against BCMA-positive, CD19-positive, and dual-positive tumor cells, including cells carrying several BCMA escape mutations.
