P-ckit-ALLO1 is an allogeneic, off-the-shelf CAR-T cell therapy developed by Poseida Therapeutics that targets the human c-kit (CD117) receptor tyrosine kinase. It is designed for dual therapeutic applications: as a non-genotoxic conditioning regimen to deplete hematopoietic stem cells (HSCs) in the bone marrow prior to transplantation and as a direct anti-tumor therapy for myeloid malignancies such as acute myeloid leukemia (AML). The therapy is manufactured using Poseida's proprietary piggyBac Gene Delivery System and Cas-CLOVER Site-specific Gene Editing System, along with a 'Booster Molecule' intended to produce a high percentage of T stem cell memory (Tscm) cells. Additionally, P-ckit-ALLO1 incorporates a fast-acting safety switch that allows for the rapid clearance of the CAR-T cells from the patient's system before the infusion of donor hematopoietic stem cells, thereby reducing potential toxicity compared to traditional chemotherapy or radiation-based conditioning.
