Overview

P-Esbp is an **HPMA-based polymer-peptide conjugate** consisting of an N-(2-hydroxypropyl)methacrylamide copolymer linked to an **E-selectin-binding peptide** with the sequence DITWDQLWDLMK. It is a **targeted macromolecular drug delivery platform** designed to bind **E-selectin** on activated endothelial cells in inflamed and angiogenic vasculature, enabling selective vascular targeting. The platform has been developed by **Vaxil Bio** under an exclusive worldwide license from **BGN Technologies** and has been explored primarily in aggressive metastatic cancer settings, especially colorectal cancer with liver metastasis, while the unconjugated platform has also shown activity in preclinical models of atherosclerosis and inflammatory disease. The more advanced therapeutic construct described in the literature is **P-Esbp-DOX**, which adds doxorubicin to the same targeting scaffold, but P-Esbp itself refers specifically to the carrier-targeting conjugate rather than the doxorubicin-containing version.
