P. acnes Avidocin is a preclinical therapeutic protein candidate developed by Pylum Biosciences for the treatment of acne. It belongs to a class of engineered R-type pyocins known as Avidocins, which are high-molecular-weight, contractile protein complexes derived from bacteriocins. The mechanism of action involves the highly specific recognition of surface receptors on *Cutibacterium acnes* (formerly *Propionibacterium acnes*), followed by the contraction of the protein sheath to punch a hole through the bacterial cell envelope. This results in rapid membrane depolarization and bacterial death. Unlike broad-spectrum antibiotics, P. acnes Avidocin is designed to be highly specific, potentially reducing the risk of antibiotic resistance and minimizing disruption to the commensal skin microbiome.
