P. falciparum 3D7 is a well-characterized, standardized clone of the *Plasmodium falciparum* parasite, originally derived from the NF54 isolate. It is utilized as a biological challenge agent in Controlled Human Malaria Infection (CHMI) studies, specifically within the CHMI-trans model developed by Radboud University Medical Center. In these clinical trials, healthy, malaria-naïve volunteers are inoculated with 3D7-infected erythrocytes to induce gametocytaemia—the sexual stage of the parasite required for mosquito transmission. This model allows researchers to evaluate the efficacy of transmission-blocking interventions (TBIs), including vaccines, monoclonal antibodies, and antimalarial drugs, by assessing the infectiousness of volunteers to *Anopheles* mosquitoes under controlled conditions. Unlike traditional therapeutics, it serves as a standardized research tool to simulate infection and transmission in a clinical environment.
