p.DOM-WT1-37 is a DNA plasmid vaccine designed for the treatment of hematological malignancies, specifically chronic myeloid leukemia (CML) and acute myeloid leukemia (AML). Developed by the University Hospital Southampton NHS Foundation Trust in collaboration with the University of Southampton, the vaccine utilizes a DNA fusion gene approach. It incorporates a sequence encoding a specific epitope (WT1-37) of the Wilms' tumor 1 (WT1) protein fused to a bacterial sequence (DOM) derived from Fragment C of tetanus toxin. This design is intended to break immune tolerance and stimulate a robust CD8+ T-cell response against leukemia cells expressing the WT1 antigen. The vaccine is typically administered via intramuscular injection followed by electroporation to enhance cellular uptake and immunogenicity, often as part of a dual-vaccination regimen alongside p.DOM-WT1-126.
