P001 is a targeted protein degrader (TPD) developed by Pamplona Biotech that targets Interleukin-1 receptor-associated kinase 4 (IRAK4). IRAK4 is a critical signaling node in the Toll-like receptor (TLR) and interleukin-1 receptor (IL-1R) pathways, which are central to the inflammatory response and the pathogenesis of various autoimmune diseases. Unlike traditional kinase inhibitors that only block the catalytic activity of IRAK4, P001 is designed to induce the degradation of the entire protein, thereby abolishing both its enzymatic and scaffolding functions. This approach potentially leads to more complete pathway inhibition and improved therapeutic outcomes. The P001 program includes an oral formulation currently in Phase 1 clinical development and a topical formulation in IND-enabling stages, aimed at treating autoimmune and inflammatory conditions.
