**P1** is an experimental colon-targeted theranostic prodrug in which 5-aminosalicylic acid is covalently linked to the fluorescent reporter 7-amino-4-methylcoumarin through an azobond. Colonic azoreductase activity reduces the azo linkage, releasing 5-aminosalicylic acid and restoring coumarin fluorescence. The molecule was investigated in polymeric micelles to improve solubility and preferential delivery to inflamed colonic tissue in a mouse model of ulcerative colitis.
