P1 is an anti-idiotype antibody vaccine candidate developed by Pomona Ricerca for the prevention of HIV-1 infection. It is specifically engineered to mimic the paratope of b12, a well-characterized human broadly neutralizing antibody (bNAb) that targets the CD4-binding site of the HIV-1 envelope glycoprotein gp120. By functioning as an 'internal image' of the HIV-1 epitope, P1 is intended to stimulate the immune system to produce antibodies with similar neutralizing breadth and potency as b12. The candidate has been developed in multiple formats, including single-chain variable fragments (scFv), minibodies, and Fab fragments, to optimize immunogenicity and protein stability. Preclinical studies in rabbit models have demonstrated that immunization with P1 can elicit a humoral response capable of neutralizing HIV-1 pseudoviruses, supporting its potential as a foundational element for an oligo-polyclonal anti-idiotype vaccine strategy.
