P10-KLH is an experimental carbohydrate-mimetic peptide (CMP) vaccine designed to induce active immunity against tumor-associated gangliosides, primarily GD2 and GD3. GD2 is a ganglioside highly expressed on tumors of neuroectodermal origin, such as melanoma, neuroblastoma, and certain breast cancers, but it typically elicits a weak immune response. P10 is a 12-amino acid peptide identified via phage display that structurally mimics the carbohydrate epitope of GD2. To enhance its immunogenicity, the peptide is covalently conjugated to the carrier protein keyhole limpet hemocyanin (KLH). When administered with an adjuvant like QS-21, P10-KLH induces the production of GD2-specific IgG antibodies and activates CD4-positive T-cell mediated delayed-type hypersensitivity (DTH) responses. A stabilized version, P10s-KLH, has been evaluated in Phase 1 clinical trials for metastatic breast cancer and melanoma to assess safety and immunogenicity.
