P110 is a cell-permeable peptide inhibitor designed to selectively disrupt the interaction between Dynamin-1-like protein (DRP1) and Mitochondrial fission 1 protein (FIS1). By specifically blocking the recruitment of DRP1 to the mitochondrial outer membrane by FIS1, P110 inhibits pathological mitochondrial fission and fragmentation without interfering with basal mitochondrial dynamics or DRP1's interaction with other receptors like MFF. This mechanism reduces mitochondrial dysfunction and the subsequent release of mitochondrial damage-associated molecular patterns (mtDAMPs). P110 has demonstrated therapeutic potential in preclinical models of Graft-versus-host disease (GVHD), where it mitigates conditioning-mediated tissue damage and pathological immune activation. Additionally, it has been extensively studied for neuroprotective and cardioprotective effects in models of Parkinson's disease, Alzheimer's disease, Huntington's disease, and ischemia-reperfusion injury.
