Drug intelligence / Profile preview
A p110β-selective inhibitor is a class of small molecule inhibitors that selectively target the p110β catalytic subunit of Class IA phosphoinositide 3-kinase (PI3K), encoded by the PIK3CB gene. Unlike pan-PI3K inhibitors, which block all four Class I PI3K isoforms (p110α, β, δ, and γ) and often cause significant off-target toxicities (such as hyperglycemia due to p110α inhibition), p110β-selective inhibitors offer a more targeted therapeutic window. They are particularly investigated for the treatment of PTEN-deficient tumors, such as glioblastoma, prostate cancer, and breast cancer, which frequently exhibit a specific dependency on p110β signaling. Additionally, p110β-selective inhibitors have been explored in other therapeutic areas, including antithrombotic therapy and Fragile X syndrome.
