P128 is a recombinant chimeric ectolysin (phage-derived lysin) with potent and rapid bactericidal activity specifically against staphylococci, including drug-resistant strains such as methicillin-resistant *Staphylococcus aureus* (MRSA) and vancomycin-resistant *S. aureus* (VRSA). It acts by cleaving the pentaglycine-bridge of peptidoglycan in the bacterial cell wall, leading to cell lysis. P128 has demonstrated efficacy both *in vitro* and *in vivo* against multidrug-resistant pathogens and is also active against coagulase-negative staphylococci (CoNS), including those present in biofilms. Its specificity allows it to kill pathogenic staphylococci without disrupting beneficial bacterial flora. Developed primarily for the treatment of serious infections caused by *S. aureus*—including nasal carriage eradication—P128 has reached Phase 2 clinical trials for indications such as bacterial infections, chronic kidney diseases with coinfection risk, and staphylococcal infections[3][4][5][6][7].
