Overview

P14-16 is an experimental immunomodulatory protein fraction, with a molecular weight of approximately 14-16 kDa, derived from *Mycobacterium bovis*. It serves as the biological component of the nanobiological response modifier OncoTherad (MRB-CFI-1). Developed by researchers at the University of Campinas (UNICAMP), P14-16 acts as an agonist for Toll-like receptors 2 and 4 (TLR2 and TLR4), thereby stimulating the innate immune system. In preclinical studies for non-muscle invasive bladder cancer (NMIBC), P14-16 has demonstrated the ability to downregulate the RANK/RANKL signaling pathway and the PD-1/PD-L1 immune checkpoint, contributing to reduced tumor progression and recurrence. It is typically administered intravesically and is studied both as a standalone agent and as a constituent of the OncoTherad complex.
