Overview

p16INK4a peptide is a synthetic peptide derived from the tumor suppressor protein p16INK4a (also known as CDKN2A), designed to mimic its activity or act as a vaccine antigen. p16INK4a naturally inhibits cyclin-dependent kinases 4 and 6 (CDK4 and CDK6), preventing RB phosphorylation and cell cycle progression from G1 to S phase. In therapeutic contexts, p16INK4a peptides have been used as: - **Direct antitumor agents:** Delivered intracellularly (often via transporter peptides), they can suppress tumor growth and induce G1 cell cycle arrest and apoptosis, as demonstrated in preclinical models of neuroblastoma, B-cell lymphoma, bladder cancer, and gliomas[1][4]. - **Vaccine candidates:** A specific p16INK4a peptide (P16_37-63) has been formulated as a peptide vaccine for patients with advanced HPV-associated, p16INK4a-overexpressing cancers.
