P1E6 is a function-blocking monoclonal antibody (IgG1) that specifically targets the alpha-2 (α2) integrin subunit (ITGA2). The α2β1 integrin is a primary receptor for type I collagen, which is highly upregulated in the desmoplastic stroma of pancreatic cancer. By blocking the interaction between tumor cells and the extracellular matrix, P1E6 inhibits collagen-mediated cell adhesion, migration, and proliferation. In preclinical studies using fluorescent orthotopic mouse models of pancreatic cancer, systemic administration of P1E6 significantly reduced primary tumor growth, prevented ascites formation, and decreased metastatic spread to organs such as the liver and spleen. These findings suggest that inhibiting α2β1 integrin-mediated interactions may have therapeutic value in treating recalcitrant, collagen-rich malignancies.
