Overview

P1G10 is a proteolytic fraction isolated from the latex of *Vasconcellea cundinamarcensis* (formerly known as *Carica candamarcensis*). It contains cysteine proteinases and has demonstrated multiple therapeutic properties including wound healing, anti-inflammatory, antioxidant, and antifungal activities. ## Therapeutic Applications P1G10 has shown significant efficacy in several clinical applications: 1. **Diabetic Foot Ulcers**: A randomized, double-blind trial with 50 patients demonstrated that topical application of 0.1% P1G10 significantly enhanced foot ulcer healing compared to hydrogel treatment. The P1G10 group showed 2.95-fold greater efficacy for complete healing and 2.52-fold higher efficacy for achieving at least 80% healing. 2. **UV Radiation Damage**: P1G10 reduces erythema after UVB exposure and demonstrates anti-inflammatory effects. 3. **Antioxidant Properties**: P1G10 reverses the depletion of antioxidant enzymes (GSH, GSH-Px, SOD) and reduces catalase activity increased by UVB exposure, leading to decreased MDA levels. 4. **Antifungal Activity**: P1G10 has demonstrated antifungal properties against *Botrytis cinerea*, showing potential as an antifungal additive in food preservation applications. ## Mechanism of Action P1G10's therapeutic effects appear to involve multiple mechanisms, primarily linked to its cysteine proteinase activity, anti-inflammatory, antioxidant, and cell cycle modulation effects. ## Safety Profile P1G10 has demonstrated a favorable safety profile in various administration routes. No systemic toxicity was observed following oral, intravenous, or subcutaneous administration, although local reactions at injection sites were noted with higher doses (phlebitis with i.v. and cutaneous lesions with s.c.). Topical application at 0.1% concentration showed no adverse effects in clinical trials. ## Formulation and Delivery P1G10 has been successfully formulated as 0.1% w/w in Polawax dressing for topical application in diabetic foot ulcer treatment. Future development may include nanoparticle formulations to enhance delivery and efficacy.
