P2-1

P2-1 is a preclinical glucose-responsive insulin (GRI) candidate originally developed by Protomer Technologies, which was acquired by Eli Lilly in 2021. The drug is engineered using Protomer's proprietary Molecular Engineering of Protein Sensors (MEPS) platform, which allows for the integration of molecular sensors directly into the structure of therapeutic proteins. In the case of P2-1, the insulin molecule is modified with sensors that respond to blood glucose levels, enabling the insulin to activate in the presence of hyperglycemia and remain inactive during periods of low glucose. This "smart insulin" approach is designed to provide automated, precise glycemic control, potentially reducing the risk of hypoglycemia and eliminating the need for frequent glucose monitoring in patients with Type 1 and Type 2 diabetes.
