Overview

P2 peptide is a synthetic mimetic peptide derived from the epitope of a detrimental anti-tetranectin (TN) monoclonal antibody, mAb9. Identified through research into the role of tetranectin in inflammatory diseases, P2 peptide was designed to repurpose a pathological epitope into a therapeutic agent. It primarily targets and neutralizes High Mobility Group Box 1 (HMGB1) and inhibits Procathepsin L, two critical drivers of dysregulated inflammation. In preclinical studies, P2 peptide has demonstrated efficacy in reducing systemic inflammation and organ failure in sepsis models, as well as alleviating joint destruction and inflammatory cytokines in rheumatoid arthritis models. By interfering with the HMGB1-mediated inflammatory cascade, it serves as a potential targeted therapeutic for conditions characterized by innate immune cell overactivation.
