Overview

P22077 is a cell-permeable, small molecule inhibitor of the deubiquitinating enzyme USP7 (Ubiquitin-Specific Protease 7), also known as HAUSP (Herpesvirus-Associated Ubiquitin-Specific Protease). It was one of the first relatively selective USP7 inhibitors identified and is widely used as a chemical probe in preclinical research. By inhibiting USP7, P22077 prevents the deubiquitination and subsequent stabilization of MDM2, leading to MDM2 degradation and the stabilization of the tumor suppressor p53. This mechanism induces p53-mediated apoptosis and growth arrest in various cancer cell lines. Research has demonstrated its potential in treating glioblastoma by targeting glioma stem cells (GSCs), as well as in neuroblastoma and other p53-dependent malignancies.
