The p24CE DNA vaccine is a therapeutic DNA vaccine designed to induce immune responses against Human Immunodeficiency Virus type 1 (HIV-1). It targets highly conserved elements (CE) within the HIV-1 p24gag protein, which are regions less prone to mutation, aiming to overcome the challenge of viral diversity in HIV-1 vaccine development. The vaccine is engineered to express these conserved elements, thereby eliciting both cellular immunity, specifically CD4+ and CD8+ T cell responses, and humoral immunity through antibody production. Preclinical studies have been conducted in mice and macaques, demonstrating its ability to induce robust immune responses. Clinical trials have evaluated its safety and immunogenicity in both healthy, HIV-uninfected adults and HIV-1 infected individuals receiving antiretroviral therapy. The vaccine is typically administered via intramuscular injection, often utilizing electroporation for enhanced delivery, and may be co-administered with adjuvants like IL-12 DNA or used in prime-boost regimens with full-length p55gag DNA.
