P25-DIV3W peptide + CD44 siRNA is a preclinical peptide-siRNA nanoparticle complex designed for the targeted treatment of HER2-positive (HER2+) breast cancer. Developed at Clemson University, the system employs a tandem peptide (P25-DIV3W) as a non-viral delivery vehicle. The P25 component serves as a targeting ligand that binds specifically to the HER2 receptor, while the DIV3W component is a fusogenic peptide that promotes endosomal escape, ensuring the intracellular delivery of the siRNA cargo. The therapeutic payload consists of siRNA targeting CD44, a cell surface glycoprotein associated with cancer stemness and tumor progression. By silencing CD44 expression, the complex significantly reduces cell viability and induces apoptosis in HER2-overexpressing cancer cells while sparing normal breast epithelial cells.
