Overview

P28z CAR T cells are second-generation chimeric antigen receptor (CAR) T cells engineered to target the prostate-specific membrane antigen (PSMA), a cell-surface glycoprotein significantly overexpressed in prostate cancer cells and the neovasculature of various solid tumors. The P28z construct features a single-chain variable fragment (scFv) derived from the J591 monoclonal antibody, which specifically binds to the extracellular domain of PSMA. This scFv is fused to a CD28 costimulatory domain and a CD3-zeta signaling domain. Upon binding to PSMA-expressing tumor cells, the CAR triggers T-cell activation, proliferation, and cytokine production, leading to targeted lysis of the malignant cells. Developed primarily at Memorial Sloan Kettering Cancer Center, P28z has been evaluated in early-phase clinical trials for patients with metastatic castration-resistant prostate cancer, serving as a foundational construct for subsequent iterations of PSMA-targeted cell therapies.
