Overview

P2Y6 shRNA is a short hairpin RNA designed to silence the expression of the P2Y purinoceptor 6 (P2Y6R), a G protein-coupled receptor that is activated by extracellular uridine diphosphate (UDP). In multiple myeloma (MM), P2Y6R is abnormally overexpressed and promotes tumor cell proliferation and survival through the Gαq/11-mediated calcium signaling pathway. By knocking down P2Y6R, this shRNA inhibits the elevation of intracellular Ca2+ levels, reduces the phosphorylation of CaMKIIγ, and suppresses the NF-κB signaling pathway. Preclinical studies conducted at Jiangxi Medical College have demonstrated that P2Y6 shRNA effectively inhibits the growth of myeloma cells in both in vitro and in vivo xenograft models, highlighting P2Y6R as a potential therapeutic target for the treatment of multiple myeloma.
