p300 siRNA is a small interfering RNA designed to silence the expression of the E1A-binding protein p300 (EP300), a histone acetyltransferase and transcriptional coactivator. In prostate cancer, p300 is frequently upregulated and serves as a critical coactivator for the androgen receptor, promoting the transcription of genes that drive tumor progression even under low-androgen conditions. By targeting p300 mRNA for degradation, this siRNA approach induces caspase-dependent apoptosis through both intrinsic and extrinsic pathways and reduces the invasive potential of cancer cells by downregulating matrix metalloproteinase-9 (MMP-9). Research indicates that p300 and its paralog CBP have distinct roles, with p300 depletion specifically triggering cell death in both androgen-dependent and castration-resistant prostate cancer models, making it a potential therapeutic strategy for advanced malignancies.
