Overview

P301 is a synthetic 15-amino acid peptide primarily utilized as a negative control in preclinical research involving Foxp3 (forkhead box P3) inhibition. In studies evaluating the therapeutic efficacy of Foxp3-inhibitory peptides like P60, P301 serves as a non-functional control to demonstrate that the observed immunomodulatory and antitumor effects are specific to the inhibition of the Foxp3 transcription factor. P301 is often derived from the HIV-1 glycoprotein gp120 (amino acids 301-315) or synthesized as a scrambled version of active peptides. While it is used in experimental models of breast cancer and other malignancies to assess the impact of regulatory T cell (Treg) blockade and direct tumor-intrinsic Foxp3 inhibition, P301 itself lacks therapeutic activity and does not inhibit Foxp3 homodimerization or nuclear translocation.
