P36-5D2 is a human monoclonal antibody isolated from individuals infected with SARS-CoV-2. It demonstrates potent and broad neutralizing activity against major SARS-CoV-2 variants of concern, including those with key mutations (K417N, E484K, N501Y) that confer resistance to other neutralizing antibodies. Structural analyses show that P36-5D2 targets a conserved epitope on the receptor-binding domain of the SARS-CoV-2 spike protein. In preclinical studies, a single intraperitoneal injection of P36-5D2 as prophylaxis completely protected animals from challenge with infectious SARS-CoV-2 Alpha and Beta variants, resulting in normal body weight and no infection-associated death up to 14 days post-challenge. Treated animals also showed significant reductions in viral load in lungs and brain and reduced lung pathology compared to controls[1][2].
