p38α MAPK-IN-1 is a potent and selective small molecule inhibitor of p38α mitogen-activated protein kinase (MAPK), also known as MAPK14, with an IC50 of approximately 0.5 nM. It is primarily utilized as a pharmacological tool in preclinical research to investigate the role of p38α signaling in various pathological conditions, including inflammatory diseases, cancer, and neurodegenerative disorders. In studies of Huntington's disease, p38α MAPK-IN-1 has been used as a reference inhibitor to demonstrate that blocking p38α phosphorylation can alleviate motor deficits and suppress neuroinflammatory pathways such as pyroptosis. It is characterized by its high potency and selectivity for the alpha isoform of p38 MAPK over other isoforms and related kinases, making it a valuable tool for validating p38α as a therapeutic target.
