Drug intelligence / Profile preview
p40-EBI3 is a novel heterodimeric cytokine composed of the interleukin-12 subunit p40 (IL-12p40) and Epstein-Barr virus-induced gene 3 (EBI3). Developed by researchers at The Catholic University of Korea, it acts as an immunoregulatory agent that promotes immune tolerance and suppresses inflammatory pathways. Mechanistically, p40-EBI3 suppresses STAT3 phosphorylation while promoting STAT5 activation, which downregulates the transcriptional repressor DREAM and restores the expression of the anti-inflammatory regulator A20. This reciprocal signaling shift leads to the expansion of regulatory T (Treg) cells and the suppression of pathogenic T helper 17 (Th17) cells, as well as the inhibition of osteoclastogenesis and necroptotic signaling. It is being investigated in preclinical models for the treatment of autoimmune and inflammatory diseases, including multiple sclerosis, rheumatoid arthritis, and systemic sclerosis.
