Overview

P5 peptide is a 21-amino acid synthetic long peptide (sequence: ELAAWCRWGFLLALLPPGIAG) derived from the rat HER2/neu protein, specifically encompassing residues 5 to 25. It is utilized as a cancer vaccine candidate designed to stimulate the immune system against HER2/neu-overexpressing malignancies. The peptide contains multiple epitopes that can be presented by both MHC class I and II molecules, thereby activating both CD8+ cytotoxic T lymphocytes and CD4+ T helper cells. In research settings, P5 is frequently pulsed onto dendritic cells and administered alongside adjuvants such as CpG-ODN and helper peptides like PADRE to enhance its immunogenicity and therapeutic efficacy by overcoming the limitations of free peptide vaccines, such as rapid degradation and weak immune stimulation.
