Overview

A synthetic peptide-based cancer vaccine composed of amino acids 264 to 272 of the wild-type human tumor suppressor protein encoded by the TP53 gene. The canonical sequence for this nonamer is LLGRNSFEV. This vaccine is designed to elicit an HLA-A2.1-restricted cytotoxic T lymphocyte (CTL) immune response against tumor cells that overexpress or accumulate mutant forms of the p53 protein—a common event in many cancers[1][5]. By presenting this specific epitope via MHC class I molecules on antigen-presenting cells or directly on tumor cells, it aims to stimulate CD8+ T cell-mediated recognition and destruction of malignant cells expressing abnormal levels or mutated forms of p53[1][3][5]. The immunogenicity and processing efficiency of this epitope can be influenced by mutations flanking the region (notably at codon 273), which may affect its presentation and subsequent immune targeting[6]. The primary indication under investigation has been various solid tumors including squamous cell carcinoma of the head and neck[3][5].
