p53-expressing oncolytic adenovirus is a preclinical-stage therapeutic candidate being developed by ORCA Therapeutics for the treatment of various cancers. This asset consists of a replication-competent adenovirus engineered to deliver and express the TP53 gene (encoding the p53 tumor suppressor protein) specifically within tumor cells. The drug employs a dual mechanism of action: direct oncolysis, where the virus replicates within and destroys cancer cells, and gene therapy-mediated restoration of the p53 pathway. By reintroducing functional p53 into p53-deficient or mutant tumors, the therapy aims to restore tumor-suppressor activities such as apoptosis and cell-cycle arrest, potentially overcoming resistance to conventional treatments and enhancing anti-tumor immune responses. It is currently being evaluated for its potential across multiple tumor types.
