p53 mRNA-LNP is an experimental therapeutic platform designed to deliver messenger RNA (mRNA) encoding the p53 tumor suppressor protein using lipid nanoparticle (LNP) technology. In many cancers, including ovarian cancer, p53 is mutated or deficient, leading to uncontrolled cell growth and resistance to conventional treatments like radiotherapy. This platform aims to restore functional p53 expression within tumor cells, thereby re-establishing apoptotic pathways and sensitizing the cells to DNA damage-induced cytotoxicity. A specific iteration of this technology, p53@HER-LNP, is functionalized with Her2-specific antibodies to enable selective targeting of Her2-overexpressing cancer cells. Preclinical studies have demonstrated that this targeted delivery significantly enhances the efficacy of radiotherapy in p53-deficient, Her2-positive models by promoting robust apoptosis and inhibiting tumor growth.
