Overview

The **p53 synthetic long peptide vaccine** is a therapeutic cancer vaccine designed to stimulate the immune system to recognize and attack tumor cells that overexpress the tumor-associated antigen (TAA) p53. The vaccine consists of multiple overlapping peptides derived from wild-type or mutant regions of the human p53 protein, typically covering amino acid positions 70–248 or 70–251. Upon administration, these peptides are processed and presented by professional antigen-presenting cells (APCs), leading to activation of both cytotoxic CD8+ T lymphocytes and helper CD4+ T cells. This dual activation aims to generate a robust anti-tumor immune response targeting cancer cells with abnormal or overexpressed p53. The approach has been evaluated in clinical trials for various cancers including ovarian cancer and metastatic colorectal cancer. Clinical studies have shown that the vaccine is generally safe and immunogenic, inducing specific T-cell responses against p53-expressing tumors[1][3][4][5][8].
