Overview

p53 TCR-NK is an investigational adoptive cell therapy consisting of natural killer (NK) cells engineered to express a T-cell receptor (TCR) specific for the p53 R175H mutation. Developed by Affini-T Therapeutics in collaboration with MD Anderson Cancer Center, this therapy targets one of the most common hotspot mutations in oncology, which is prevalent in various solid tumors including colorectal, ovarian, and lung cancers. By utilizing NK cells as the effector platform, the therapy aims to leverage their innate anti-tumor properties and lower risk of graft-versus-host disease (GvHD) compared to traditional T-cell therapies. The engineered TCR allows the NK cells to precisely recognize the p53 R175H neoantigen presented by HLA-A*02:01 on the surface of malignant cells, triggering targeted cytotoxicity. The program is currently in the preclinical stages of development.
