Overview

p53MVA is a cancer vaccine based on a recombinant Modified Vaccinia Ankara (MVA) viral vector engineered to express the full-length wild-type human tumor suppressor protein p53. The primary mechanism involves delivering the wild-type human p53 gene to antigen-presenting cells via the MVA vector, which leads to expression and presentation of multiple antigenic determinants of p53 on different HLA molecules. This process aims to stimulate robust CD8+ T cell responses against tumors that overexpress or present mutated forms of p53. Preclinical and early clinical studies have shown that immunization with p53MVA can induce anti-tumor immunity and activate cytolytic T cells specific for wild-type human p53 epitopes. The vaccine has been evaluated alone and in combination with immune checkpoint inhibitors such as pembrolizumab in patients with advanced solid tumors including gastrointestinal cancers, breast cancer, pancreatic cancer, hepatocellular carcinoma, head and neck cancer, ovarian cancer, primary peritoneal cancer, fallopian tube cancer, and other solid tumors[1][2][3][4][5].
