Overview

P53sh-MSC-derived exosomes are extracellular vesicles isolated from the conditioned media of bone marrow-derived mesenchymal stromal cells (BM-MSCs) that have been genetically modified to silence the p53 (TP53) gene using short hairpin RNA (shRNA). Developed in the laboratory of Sabyasachi Sen, these exosomes are being investigated as a cell-free therapeutic modality for treating obesity and type 2 diabetes mellitus (T2DM). The silencing of p53 in the parent MSCs alters the cargo of the secreted exosomes, which, when applied to target cells, has been shown to inhibit adipogenesis and reduce oxidative stress through the upregulation of antioxidant pathways. Preliminary research indicates potential applications in managing white adipose tissue hyperplasia and non-alcoholic fatty liver disease (NAFLD).
