The p55Gag plasmid DNA vaccine is a DNA-based vaccine that expresses the full-length HIV-1 HXB2 Gag protein from the human cytomegalovirus (huCMV) promoter followed by the BGH polyA signal. It has been tested in clinical trials, including HVTN 119 (NCT03181789), as both a standalone vaccine and in prime-boost regimens. The vaccine is designed to induce T cell responses against HIV-1 Gag proteins, which have been associated with better control of HIV viremia. When used in combination with conserved elements (CE) vaccines, it has shown the ability to increase breadth and epitope recognition throughout p55Gag compared to p55Gag DNA alone.
