Overview

P57AS3 is an oxypregnane steroidal glycoside isolated from the African plant Hoodia gordonii, traditionally used for its appetite suppressant properties. It is believed to be the primary active constituent responsible for the anorexigenic (appetite-suppressing) effects of Hoodia extracts. Mechanistically, P57AS3 acts centrally in the brain, increasing ATP content in hypothalamic neurons by 50–150%, which is associated with reduced food intake in animal models. Unlike cardiac glycosides with similar steroidal cores, it does not bind or alter activity of known receptors such as Na/K ATPase. In vitro studies show that it is metabolically stable and exhibits weak inhibition of CYP3A4 but not other major cytochrome P450 enzymes. Its intestinal transport involves efflux via multidrug resistance-associated proteins (MRP1/MRP2) and P-glycoprotein (P-gp). The compound has been investigated primarily for obesity and weight loss[1][2][4][6].
