P60 is a synthetic 15-mer peptide designed to inhibit the function of Forkhead box P3 (Foxp3), a transcription factor essential for the development and suppressive function of regulatory T cells (Tregs). In the tumor microenvironment, Tregs impair the efficacy of cancer immunotherapies by suppressing antitumor immune responses. P60 enters cells and binds to Foxp3, interfering with its nuclear translocation and its ability to associate with transcription factors such as NF-κB and NFAT. Beyond its effect on Tregs, P60 has been shown to directly inhibit Foxp3-expressing tumor cells, reducing the secretion of immunosuppressive cytokines like IL-10. Developed largely within academic research settings, it has demonstrated potential as both a monotherapy and an adjuvant to boost the efficacy of dendritic cell vaccines in preclinical models of breast cancer.
