p65 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the p65 (RelA) subunit of the NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) transcription factor. By utilizing the RNA interference (RNAi) pathway, it leads to the sequence-specific degradation of p65 mRNA, thereby reducing the levels of the p65 protein. This suppression inhibits the activation of the NF-κB signaling pathway, which is a central regulator of inflammatory responses, immune cell activation, and cell survival. In preclinical research, p65 siRNA has been explored as a potential therapeutic for various inflammatory diseases, such as rheumatoid arthritis and acute lung injury, as well as cancers like glioblastoma and colon cancer. It is frequently studied in conjunction with specialized delivery systems, including lipid nanoparticles, micelles, and DNA nanostructures, to overcome the challenges of oligonucleotide delivery.
