Overview

P667 is a synthetic chimeric decapeptide derived from Luteinizing Hormone-Releasing Hormone (LHRH). It is designed as a component of therapeutic vaccines intended for the treatment of hormone-dependent cancers, particularly advanced prostate cancer. The peptide is typically constructed by linking an LHRH-derived sequence to a T-helper cell epitope to enhance its immunogenicity and overcome self-tolerance to the endogenous hormone. When administered with an adjuvant like Montanide ISA50, P667 induces the immune system to produce high titers of antibodies that specifically neutralize endogenous LHRH. This neutralization prevents LHRH from stimulating the pituitary gland to release gonadotropins, thereby suppressing the production of testosterone to castrate levels (a process known as immunocastration). This therapeutic approach aims to provide a long-lasting alternative to surgical castration or continuous treatment with LHRH agonists or antagonists. Development of this peptide vaccine has been primarily led by the National Health Research Institutes (NHRI) in Taiwan.
