Drug intelligence / Profile preview
P7N4-Pen is a synthetic peptide derived from a functional motif of Signaling Lymphocytic Activation Molecule Family 1 (SLAMF1) with a single amino acid substitution (Y4N) and linked to the cell-penetrating peptide Penetratin (Pen) for intracellular delivery. Developed by the Norwegian University of Science and Technology (NTNU) and St. Olavs Hospital, P7N4-Pen is being investigated as a novel therapeutic candidate for the treatment of multiple myeloma (MM). Mechanistically, P7N4-Pen directly binds to and targets the oncogenic transcription factor Interferon Regulatory Factor 4 (IRF4) for degradation. This direct interaction disrupts key pro-survival pathways in MM cells, leading to the downregulation of downstream oncogenes such as MYC and β-catenin, and the inhibition of Akt and ERK1/2 phosphorylation, ultimately inducing both extrinsic and intrinsic apoptosis. In preclinical studies, P7N4-Pen has demonstrated potent anti-cancer activity against various human myeloma cell lines, including those resistant to proteasome inhibitors, and primary CD138+ patient-derived tumor cells, while sparing healthy peripheral blood mononuclear cells. It also exhibits synergistic effects when combined with chemotherapeutic agents like melphalan or proteasome inhibitors like bortezomib.
