p95HER2 CAR-T cells are an investigational adoptive cell therapy consisting of autologous T cells engineered to express a chimeric antigen receptor (CAR) specifically targeting p95HER2. p95HER2 is a truncated form of the Human Epidermal Growth Factor Receptor 2 (HER2) that lacks the extracellular domain, a modification that renders tumors resistant to standard HER2-targeted therapies like trastuzumab. This truncated protein is expressed in approximately 40% of HER2-positive tumors but is absent in healthy tissues, providing a highly selective target for immunotherapy. Developed by the Vall d'Hebron Institute of Oncology (VHIO), this therapy is currently being evaluated in Phase 1 clinical trials for patients with advanced p95HER2-expressing solid tumors, including breast, gastric, and endometrial cancers, who have progressed on standard treatments.
