p95HER2 LINK CAR T cells are an investigational, logic-gated chimeric antigen receptor (CAR) T-cell therapy designed for the treatment of HER2-positive solid tumors. Developed by the Vall d'Hebron Institute of Oncology (VHIO) and the Hospital del Mar Research Institute, this therapy utilizes the LINK CAR platform to selectively target cells co-expressing both the tumor-specific truncated HER2 variant, p95HER2, and full-length HER2. The LINK CAR design splits T-cell activation signaling between two separate CAR constructs: one incorporating the LAT signaling domain and the other incorporating the SLP-76 signaling domain. This Boolean AND-gate logic ensures that the CAR T cells are only activated in the presence of both antigens, thereby sparing normal tissues that express only basal levels of HER2 and mitigating on-target, off-tumor toxicities. The therapy has demonstrated preclinical efficacy and safety in in vitro and in vivo models of breast, gastric, ovarian, and lung cancers.
