Drug intelligence / Profile preview
**p95HER2.CAR T cells** are an investigational autologous, second-generation CAR-T-cell therapy originating at Vall d'Hebron Institute of Oncology for HER2-amplified solid tumors expressing p95HER2, a tumor-specific truncated HER2 isoform. Patient T cells are engineered with a CAR incorporating a p95HER2-binding single-chain variable fragment, a CD8 hinge, and CD28/CD3-zeta signaling domains. The CAR is designed to selectively eliminate p95HER2-positive tumor cells while sparing normal cells that express physiologic levels of full-length HER2. Preclinical studies showed activity in breast-cancer cell-line, metastatic, and patient-derived xenograft models, although the unarmed product had more limited activity in patient-derived xenografts than the separate next-generation p95HER2.CAR-TECH2Me T-cell product.
