PA-161 is a first-in-class HER2-targeted sialidase conjugate developed by Palleon Pharmaceuticals using its proprietary EAGLE (Enzyme-Antibody Glycan Leveraging Engine) platform. The therapeutic consists of a human sialidase enzyme fused to a HER2-targeting monoclonal antibody. PA-161 is designed to treat HER2-positive solid tumors by selectively desialylating the tumor cell surface. Sialic acids are terminal sugars on cell-surface glycans that function as a "glyco-immune checkpoint" by binding to inhibitory Siglec receptors (such as Siglec-7 and Siglec-9) on various immune cells, including Natural Killer (NK) cells, macrophages, and T cells. By removing these sialic acid residues, PA-161 disrupts the immunosuppressive Siglec signaling pathway, thereby enhancing the innate and adaptive immune response against the tumor. This approach aims to overcome resistance to existing HER2-targeted therapies by modulating the glycan-mediated immune evasion mechanism.
